    Benchmark Calculations for Perchlorate from Three Human Cohorts

    The presence of low concentrations of perchlorate in some drinking water sources has led to concern regarding potential effects on the thyroid. In a recently published report, the National Academy of Sciences indicated that the perchlorate dose required to cause hypothyroidism in adults would probably be > 0.40 mg/kg-day for months or longer. In this study, we calculated benchmark doses for perchlorate from thyroid-stimulating hormone (TSH) and free thyroxine (T(4)) serum indicators from two occupational cohorts with long-term exposure to perchlorate, and from a clinical study of volunteers exposed to perchlorate for 2 weeks. The benchmark dose for a particular serum indicator was defined as the dose predicted to cause an additional 5 or 10% of persons to have a serum measurement outside of the normal range. Using the data from the clinical study, we estimated the half-life of perchlorate in serum at 7.5 hr and the volume of distribution at 0.34 L/kg. Using these estimates and measurements of perchlorate in serum or urine, doses in the occupational cohorts were estimated and used in benchmark calculations. Because none of the three studies found a significant effect of perchlorate on TSH or free T(4), all of the benchmark dose estimates were indistinguishable from infinity. The lower 95% statistical confidence limits on benchmark doses estimated from a combined analysis of the two occupational studies ranged from 0.21 to 0.56 mg/kg-day for free T(4) index and from 0.36 to 0.92 mg/kg-day for TSH.     High-resolution 3D weld toe stress analysis and ACPD method for weld toe fatigue crack initiation

    Weld toe fatigue crack initiation is highly dependent on the local weld toe stress-concentrating geometry including any inherent flaws. These flaws are responsible for premature fatigue crack initiation (FCI) and must be minimised to maximise the fatigue life of a welded joint. In this work, a data-rich methodology has been developed to capture the true weld toe geometry and resulting local weld toe stress-field and relate this to the FCI life of a steel arc-welded joint. To obtain FCI lives, interrupted fatigue test was performed on the welded joint monitored by a novel multi-probe array of alternating current potential drop (ACPD) probes across the weld toe. This setup enabled the FCI sites to be located and the FCI life to be determined and gave an indication of early fatigue crack propagation rates. To understand fully the local weld toe stress-field, high-resolution (5 mu m) 3D linear-elastic finite element (FE) models were generated from X-ray micro-computed tomography (mu-CT) of each weld toe after fatigue testing. From these models, approximately 202 stress concentration factors (SCFs) were computed for every 1 mm of weld toe.     Clinical manifestations of human brucellosis : a systematic review and meta-analysis

    BACKGROUND: The objectives of this systematic review, commissioned by WHO, were to assess the frequency and severity of clinical manifestations of human brucellosis, in view of specifying a disability weight for a DALY calculation. METHODS/PRINCIPAL FINDINGS: Thirty three databases were searched, with 2,385 articles published between January 1990-June 2010 identified as relating to human brucellosis. Fifty-seven studies were of sufficient quality for data extraction. Pooled proportions of cases with specific clinical manifestations were stratified by age category and sex and analysed using generalized linear mixed models. Data relating to duration of illness and risk factors were also extracted. Severe complications of brucellosis infection were not rare, with 1 case of endocarditis and 4 neurological cases per 100 patients. One in 10 men suffered from epididymo-orchitis. Debilitating conditions such as arthralgia, myalgia and back pain affected around half of the patients (65%, 47% and 45%, respectively). Given that 78% patients had fever, brucellosis poses a diagnostic challenge in malaria-endemic areas. Significant delays in appropriate diagnosis and treatment were the result of health service inadequacies and socioeconomic factors. Based on disability weights from the 2004 Global Burden of Disease Study, a disability weight of 0.150 is proposed as the first informed estimate for chronic, localised brucellosis and 0.190 for acute brucellosis. CONCLUSIONS: This systematic review adds to the understanding of the global burden of brucellosis, one of the most common zoonoses worldwide. The severe, debilitating, and chronic impact of brucellosis is highlighted. Well designed epidemiological studies from regions lacking in data would allow a more complete understanding of the clinical manifestations of disease and exposure risks, and provide further evidence for policy-makers.     Characterization of indoor/outdoor settled dust and air pollutants in Damaturu, Nigeria

    This paper presents results of on-going experiments being undertaken to characterise the composition of settled dusts and some airborne contaminants in homes and outdoors in Damaturu, north-eastern Nigeria. These have involved the physical analysis of the dust samples collected from different homes by scanning electron microscopy (SEM) which showed images of particles of 2µm to 50µm and fragmented filamentous particles of 50µm to 200µm. Microbiological analysis conducted include media culture and microscopic observations where a range of fungal and bacterial colonies were identified with colony forming units in settled dust and air of between 1.02 10 4 to1.18 10 5 CFU g -1 and 1.07 10 4 1.69 10 5 CFU m -3 (for fungi)and 7.70 10 3 to1.14 10 5 CFU g -1 , 1.90 10 3 to 1.13 10 5 CFU m -3 (for bacteria) . Metal analysis by ICP-MS (total quant method) of the settled dusts revealed the presence of metal elements in the range of Al>Na>Mg>Ca>K>P>Si>Mn>Ti and indicated the maximum concentration of Lead, Copper, Cadmium, Nickel and Zinc (mg kg -1 ) to bePb-89.7, Cu-68.4, Cd-15.6, Ni-14.9, Zn-1.15.The results also revealed the presence of organic chemical pollutants in varying concentrations in the indoor and outdoor dusts and airborne samples within different homes; eight semi-volatile and volatile organic compounds(SVOCs/VOCs); benzene, hex anal, nonanal, diethyl phthalate (DEP), diisobutylphthalate (DIBP), dibutylphthalate (DBP), and diethylhexylphthalate (DEHP) and limonene were quantified among the numerous organic chemicals present in the samples analysed based on their frequency of occurrence in the samples collected, as well as their potential health effects arising from short term and long term exposure as reported in earlier studies. The results suggested that the indoor environments contain considerably more organic chemical pollutants than the outdoors. From the results obtained so far on the chemical, metal, and biological pollutants present as well as the physical characteristics of indoor dusts from Nigerian homes, it can be ascertained that the dusts act as a reservoir of pollutants that can increase peoples’ exposure and be a risk to the health of residents of the community

    Microbial gene abundance and expression patterns across a river to ocean salinity gradient

The definitive version was published in PLoS One 10 (2015): e0140578, doi: 10.1371/journal.pone.0140578.Microbial communities mediate the biogeochemical cycles that drive ecosystems, and it is important to understand how these communities are affected by changing environmental conditions, especially in complex coastal zones. As fresh and marine waters mix in estuaries and river plumes, the salinity, temperature, and nutrient gradients that are generated strongly influence bacterioplankton community structure, yet, a parallel change in functional diversity has not been described. Metagenomic and metatranscriptomic analyses were conducted on five water samples spanning the salinity gradient of the Columbia River coastal margin, including river, estuary, plume, and ocean, in August 2010. Samples were pre-filtered through 3 μm filters and collected on 0.2 μm filters, thus results were focused on changes among free-living microbial communities. Results from metagenomic 16S rRNA sequences showed taxonomically distinct bacterial communities in river, estuary, and coastal ocean. Despite the strong salinity gradient observed over sampling locations (0 to 33), the functional gene profiles in the metagenomes were very similar from river to ocean with an average similarity of 82%. The metatranscriptomes, however, had an average similarity of 31%. Although differences were few among the metagenomes, we observed a change from river to ocean in the abundance of genes encoding for catabolic pathways, osmoregulators, and metal transporters. Additionally, genes specifying both bacterial oxygenic and anoxygenic photosynthesis were abundant and expressed in the estuary and plume. Denitrification genes were found throughout the Columbia River coastal margin, and most highly expressed in the estuary. Across a river to ocean gradient, the free-living microbial community followed three different patterns of diversity: 1) the taxonomy of the community changed strongly with salinity, 2) metabolic potential was highly similar across samples, with few differences in functional gene abundance from river to ocean, and 3) gene expression was highly variable and generally was independent of changes in salinity.This study was carried out within the context of the Science and Technology Center for Coastal Margin Observation & Prediction (CMOP) supported by the National Science Foundation, grant number OCE-0424602 to Antonio Baptista (http://www.stccmop.org)
